چکیده

the binding of proportionately larger quantities of the third component of complement (C3) to these cells. The same amount of bound C3 was found on the membranes of normal and HEMPAS cells for a given degree of lysis. Hence, the marked increase in lysis of HEMPAS cells is due to the increased adsorption of antibody and/or increased binding of C4. PNH cells bound the same amount of C4 per bound C1 as normal cells but bound more C3 than normal cells. However, the mean concentration of C3 on the membrane of PNH cells was one-third to one-fifth that on normal cells for a given degree of lysis. Hence, the increased lysis of PNH cells is due to the increased binding of C3 and increased hemolytic effectiveness of the bound C3. INTRODUCTION Normal red cells are not readily lysed by cold agglutinins and complement; lysis usually occurs only wheoi relatively large amounts of antibody and/or complement are used. The red cells in at least two blood disorders appear to be more readily lysed than normal red cells by anti-I cold agglutinins and complement. The red cells of patients with paroxysmal nocturnal hemoglobinuria (PNH)1 have been extensively studied and have been shown to consist of at least two populations, one of which is markedly sensitive to the lytic action of complement (1, 2). This sensitivity is apparent when complement is activated either by antibody or by the alternate (properdin) pathway (3-6). The latter mechanism of activation is responsible for the lysis of PNH cells in acidified normal serum (3, 4). Crookston et al. reported that the red cells of patients with a form of congenital dyserythropoietic anemia characterized by multinucleated erythroblasts, are also lysed in acidified normal serum (7). They proposed the name hereditary erythroblastic multinuclearity with a positive acidified serum test, or HEMPAS; this 1Abbreviations used in this paper: ACD, acid-citratedextrose solution; Cl, C2, C3.. ., the first, second, third, etc. components of serum complement (a bar over the number indicates that the component is in its active form); CLS H5D, a unit of sensitivity to complement lysis, viz. the reciprocal of the dilution of complement 0.2 ml of which will lyse 50%yo of 2.2 X 10' sensitized red cells in a total volume of 1.5 ml; CoF, a factor derived from cobra venom which initiates the alternate pathway of C3 activation; EAC, red cells sensitized with antibody and coated with components of complement; HEMPAS, hereditary erythroblastic multinuclearity with a positive acidified serum test; PNH, paroxysmal nocturnal hemoglobinuria; VBS, veranol-buffered saline. The immunomodulatory effects of shark cartilage on the mouse and human immune system

Background: Sharks get cancer rarely. A major difference between these animals and other species is that sharks have a great amount of cartilaginous tissue. Immunomodulatory effects of the cartilage of some species (cow) have been proved. Because the immune system has a major role in the defense of the body against cancer, we studied the effects of shark cartilage on the mouse and human immune...

Red Cell Enzymopathies in Patients with Hemolytic Anemia in Southern Iran: Case Series

Abstract Background: Hereditary red cell enzyme disorders are a group of Non-immune/Spherocytic Hemolytic Anemia, although these disorders are rare and they have not public health problems, the detection of these defects could help to physician in treatment and differential diagnosis. This study evaluated 5 enzymopathies in patients with Hereditary Non –immune/Spherocytic Hemolyti...

Mechanisms of immune lysis of the red cells in hereditary erythroblastic multinuclearity with a positive acidified serum test and paroxysmal nocturnal hemoglobinuria.

The red cells of patients with hereditary erythroblastic multinuclearity with a positive acidified serum test (HEMPAS), a form of congenital dyserythropoietic anemia, and the cells of patients with paroxysmal nocturnal hemoglobinuria (PNH) are lysed more readily than normal cells by certain antibodies, notably cold agglutinins (anti-I) and complement. With some but not other examples of anti-I,...

ABSTRACT: The effect of protein deficiency on the immune response upon primary and secondary stimulation with 5×108 sheep red blood cells (SRBC) was studied in 4 groups of mice fed by diets containing 4 or 21 percent of protein.Primary and secondary immune response were assessed by quantification of direct and indirect plaque forming cells (PFC) in immune spleens and by titration of serum hema...

Prevalence and Haemolytic Significance of Red Cell Antibodies among Dangerous Universal Donors in a Tertiary Care Hospital in South India

Background and Aims: A subgroup of group O individuals called ‘dangerous universal donors’ have immune (IgG) anti A and anti B antibodies which are active at 37˚C and capable of reacting with the red cells and causing lysis. The aim of this study was to find the prevalence of dangerous O group among the voluntary donor population and to assess the relation between the degree of haemolysis and t...

New insight into the immunomodulatory mechanisms of Tretinoin in NMRI mice

Objective(s):Recent evidence have proposed that Tretinoin produced in the gut preferentially promote differentiation of FoxP3+Treg cells but inhibits Th17 lymphocytes, and this may be the main immunomdulatory mechanism of Tretinoin  in vivo. This study was done to investigate the effects of Tretinoin in outbred white mice after challenge with sheep red blood cells (SRBC). Materials and Methods:...
